Well there is a few strategies, but right now its matchweek 5 and only Man Utd are guaranteed through if I remember correctly. So I think most other teams other than Man Utd should still be putting out their strongest teams... therefore not necessary to use yet. You could use it on matchday 6 if there are more top teams guaranteed progress and therefore will rest players and you need to transfer more than 1 or 2 players. Or if you don't need to use it yet, then for the later knockout stages, if you've fully backed one team or two and they both happen to get kicked out, then 6 transfers may not be enough to field a full team and you need to use wildcard...
